Cell-cycle analysis, growth factors and proliferative activity in middle ear cholesteatoma

چکیده
In order to provide much more understanding into the proliferation and differentiation of the epithelia in cholesteatoma, cell cycle associated molecules were studied by anti-PCNA and anti-Ki67, the two well characterized antibodies. In addition, The experiments included expression, localization, and quantification of EGF, TGF, EGF-R in the cholesteatoma. This study demonstrates the presence of TGFand EGF-R in the epithelium and TGF, EGF and EGF-R in the stroma of cholesteatoma. As cholesteatoma is a disease with progressive growth of the epithelium, our result showed a high MIB 1/PCNA index means the importance of the inflamed stroma. Taking all the results showing in our study, it may draw a conclusion that TGF, EGF, and EGF-R are involved in the formal pathogenesis of cholesteatoma. These results might help us to define the epithelial cells behavior in cholesteatoma clinically.

Identification of specific gene expression profiles in fibroblasts derived from middle ear cholesteatoma.
OBJECTIVE To investigate the role of fibroblasts in the pathogenesis of cholesteatoma. DESIGN Tissue specimens were obtained from our patients. Middle ear cholesteatoma-derived fibroblasts (MECFs) and postauricular skin-derived fibroblasts (SFs) as controls were then cultured for a few weeks. These fibroblasts were stimulated with interleukin (IL) 1alpha and/or IL-1beta before gene expression...
متن کاملCYTOKINE AND GROWTH FACTOR MODULATION OF CELL CYCLE EVENTS IN HUMAN MELANOMA CELL LINES
Cytokines influence cell cycle events, which in some but not all instances can be associated with melanoma progression. Analysis of the G0/G 1 and S phase fractions of the cell cycle was used to assay the proliferative or inhibitory activity of cytokines against ten human melanoma cell lines, including pairs of cell lines derived from primary and metastatic tissue of individual patients. Cy...

HYPOTHESIS This study aimed to analyze the localization and distribution of vessels and of these angiogenic growth factors: basic fibroblast growth factor (FGF-2), transforming growth factor-alpha (TGF-alpha), transforming growth factor-beta1 (TGF-beta1), and vascular endothelial growth factor (VEGF) in middle ear cholesteatoma in comparison with normal middle ear mucosa and auditory meatal ski...
متن کاملComparative Analysis of the Proliferative Activity of Cholesteatoma Epithelium
BACKGROUND: The histopathological characteristics of cholesteatoma are hyperproliferation and differentiation of the epithelium. The proliferative capacity of cholesteatoma was studied through the kinetics of the epithelium of cholesteatoma. The present study was undertaken to compare the rate of proliferation and apoptosis using immunohistochemistry with PCNA antigen and p53 protein in cholest...

Introduction: First branchial cleft anomalies manifest with duplication of the external auditory canal. Case Report: This report features a rare case of microtia and congenital middle ear and canal cholesteatoma with first branchial fistula. External auditory canal stenosis was complicated by middle ear and external canal cholesteatoma, but branchial fistula, opening in the zygomatic root and...
